NanoLab (VHH Developability)
Model

NanoLab is Lilly TuneLab's developability model for VHH domains — camelid single-domain antibodies, commonly known as nanobodies. It predicts five biophysical developability endpoints directly from a VHH heavy-chain sequence.
It is built on a state-of-the-art protein language model (ESMC) by BioHub¹ and trained on a purpose-built VHH developability dataset generated by a partner BigHat Biosciences. It is designed for teams working with single-domain formats who need to rank and prioritize candidates during lead optimization, before committing to wet lab characterization.

What does the model provide?
  • Five developability predictions from sequence: apparent molecular weight by CE-SDS, protein concentration and total yield after purification, A260/A280 absorbance ratio, and SEC monomer purity.

  • A conformal prediction interval on every endpoint, making per-sequence uncertainty explicit.

Technical specifications
Modality
VHH / single domain (nanobody).
Input format
VHH heavy-chain amino acid sequence, one per row. Canonical twenty amino acids; typical VHH length 105–135 residues.
Output format
A point estimate, plus the lower and upper bound of the conformal prediction interval.
Training data type
A purpose-built VHH developability dataset generated by a partner biologics platform (BigHat Biosciences) — approximately 2500 VHH sequences characterized across the five endpoints.
Predicted endpoint(s)
Apparent molecular weight (CE-SDS, kDa), A260/A280 absorbance ratio, protein concentration (ng/µL), total protein yield (ng), SEC monomer purity (% of total).

FAQs
How should I use the predictions?
To rank and prioritize candidates relative to one another during lead optimization. Predictions are advisory; wet lab confirmation remains appropriate to inform decisions.
What do the intervals mean?
Each endpoint returns a point prediction plus a lower and upper bound derived from conformal calibration. A wide interval indicates the molecule is expected to have larger-than-usual model errors and the point prediction should be treated with caution.
Can I use it on full-length antibodies?
No. NanoLab covers single-domain VHH only. For paired heavy-and-light-chain formats use AbLab.
Will it run on sequences outside the typical VHH length range?
Yes, the model accepts them, but they are likely to be outside the training distribution.
